Abstract:
Water samples were collected at bottom, mid-depth, and surface at six stations along a transect running from the middle of Mobile Bay to the shelf in the northern Gulf of Mexico, from June 2010 to May 2012. Data include concentrations of dimethyl sulfide (DMS), dimethylsulfoniopropionate (DMSP), dissolved oxygen, and dissolved methane, and bacterial biomass production via leucine incorporation. The transect utilized for this study is part of the Fisheries Oceanography of Coastal Alabama (FOCAL) program, which collects long-term baseline hydrographic data and plankton samples toward understanding the highly variable nature of Alabama's nearshore environment as it relates to fisheries production.
Purpose:
The purpose of this study was to understand carbon and oxygen dynamics on the Alabama shelf in relation to potential inputs of hydrocarbons from the BP-Macondo spill.

Water samples were collected at bottom, mid-depth, and surface approximately once per month from June 15 2010 to May 15 2012 at 6 stations on the FOCAL transect running from the middle of Mobile Bay to the shelf in the northern Gulf of Mexico. Samples were collected with Niskin bottles on a CTD rosette. Intervals between sampling ranged from 2 to 6 weeks. Surface samples were always collected, but sometimes the mid-depth and occasionally the bottom samples were omitted. Most sampling events included all six stations.

Dimethyl sulfide (DMS) concentrations: DMS concentrations measured by pure-and-trap gas chromatography with flame photometric detection (FPD). Two to four ml of seawater were purged with helium and the DMS cryotrapped in Teflon tubing. DMS introduced into Chromosil 330 column and quantified with FPD. Dissolved methane concentrations: Methane was quantified with a headspace equilibration technique and GC-FID detection. Water samples were filled without gas exchange into 160 ml serum bottles. Immediately after filling, bottles were sealed with a rubber stopper leaving no headspace. Samples were stored at in situ temperature until analysis in the shore laboratory. To each bottle, 4 ml of pure N2 gas was introduced with a glass barreled syringe and 22 gage needle, allowing water displacement. The bottle was shaken vigorously to equilibrate liquid and gas phases. The headspace was sampled for CH4 concentration by GC-FID and the original dissolved CH4 concentration was obtained from the total methane in the bottle, accounting for gas and liquid phases and the temperature and salinity for that sample. Precision was 5% or better.

Bacterial biomass production via leucine incorporation: Bacterial production was measured by tritiated (3H) leucine incorporation into TCA-insoluble macromolecules. The microcentrifuge method was used. Triplicate live samples (2 ml) and one TCA-killed sample per depth were treated with 20 nM of 3H-Leucine. Incubation was for 1 h and samples were killed by addition of TCA. Samples were spun at 15,000 x g in a microcentrifuge and after several rinses, the pellet was resuspended in Ecolumn scintillation cocktail and the 3H radioactivity quantified. Rates are reported in picomolar Leucine per hour. Precision is generally 10%. (Smith, D. C., & Azam, F. (1992).
