Abstract:
Abundances of viruses, prokaryotes, diatoms, dinoflagellates, ciliates and heterotrophic nanoflagellates were determined over time in mesocosm experiments measuring the effects of oil, dispersant and dispersed oil on the microbial loop. Two separate experiments were carried out in June and August 2011. Abundances in the treated mesocosms were compared to a no addition control and a glucose addition control.
Purpose:
These experiments were designed to quantify the effects of oil, dispersant and dispersed oil on the coastal pelagic microbial food web.

Replicated mesocosms (n=5) were exposed to different treatments (control, glucose, oil, dispersant and dispersed oil) and sampled over time for biomass of different microbial groups.

Abundances: At specific times, 4.5 ml samples were collected from each mesocosm (from t=0 to t=5 days) and fixed in 0.5% EM grade glutaraldehyde and flash frozen in liquid nitrogen. Samples were stored at -80C until processing. Samples for viruses were diluted 100x in filtered TE and stained with Sybr Green I at 80C for 10 min. Samples were then run for 2 min on a FACSCalibur flow cytometer with calibrated flow rate to determine abundances. For prokaryotes, dilution was 10X with TE, and samples were stained for 10 min at room temperature before analysis on the flow cytometer. For heterotrophic nanoflagellates, 50 ml samples were fixed in 0.5% EM grade glutaraldehyde for 30 min before being filtered through a 0.8 micron black polycarbonate filter. Cells were stained with DAPI and counted using epifluorescence microscopy. For phytoplankton and ciliates, ~100 ml was fixed in acidic Lugol's solution and counted using an inverted microscope.

Growth rates and virus production rates: At t=0, 1 and 3 days, sub-samples were collected from each mesocosm and 5 ml of water was added to 45 ml of 30 kDa filtered water. Sub-samples were collected at t=0, 3, and 6 hours and the abundance of prokaryotes and viruses determined. Growth and viral production rates were determined based on the regression of ln(cell or virus number) vs time over 6 hours.
