Abstract:
Nutrients and water quality have been measured in three lagoonal environments in Perdido Bay, Florida, since Dec 2000. The data include dissolved inorganic nitrogen, dissolved organic nitrogen, phosphate, water column chlorophyll, benthic chlorophyll, and particulate organic matter (POM).
Purpose:
To improve our understanding of lagoonal environments, and to improve our ability to create effective policies of environmentally-sustainable development and use of coastal watersheds by humans. The data are part of an ongoing survey of the physical, chemical and biological variables of three distinct lagoonal environments in Perdido Bay, Florida. The three lagoons cover a wide gradient of human pressure, from a pristine lagoon to a lagoon with a moderately developed watershed to a lagoon with a highly developed watershed, thereby allowing us to make inferences as to how human utilization of watersheds affects the health of coastal lagoons and services offered to humankind and wildlife.

Process_Description:
Discrete water samples were taken at the following points within each lagoon: near the mouth, near the upstream edge, on both sides of the lagoon, and at two additional random points. Dissolved nitrogen species were determined using colorimetric techniques on a Skalar autoanalyzer. Particulate organic matter (POM) was calculated by loss on ignition (LOI). | For benthic chlorophyll, we sampled the sediment using a 2.5 cm diameter corer and sliced the top 1 cm sediment layer into a centrifuge tube, which was stored on ice and transported to the lab. In the lab, samples were frozen at -80 degrees C until processed. Upon thawing, chlorophyll was extracted for a 24 h period with 25.0 ml of acetone (Strickland JDH, Parsons TR. 1972. A practical handbook of seawater analysis, 2nd edn. Bulletin of the Fisheries Research Board of Canada 167: 1-310). Chl a concentrations were measured fluorometrically with a Turner Designs Model TD-700 fluorometer using acidification (1N HCl) to correct for the presence of phaeopigments for samples collected before 2010, and with the Welshmeyer protocol extraction (Welschmeyer, NA. 1994. Fluorometric analysis of chlorophyll a in the presence of chlorophyll b and pheopigments. Limnology and Oceanography 39(8): 1985-1992) for samples collected in 2010 and later.
