- A transect was sampled from Mobile Bay, Alabama to 40 km offshore at monthly to bi-monthly intervals from March, 2007. Horizontal salinity gradients ranged from 8 to 18 PSU in surface waters. Temperature ranged from 16 to 32 C. Maxima in chlorophyll concentrations and total microalgae numbers were associated with maxima in nutrient concentration. Nutrient concentrations were in turn inversely correlated with salinity, indicating the importance of Mobile Bay as a source. The highest chlorophyll concentrations were found in July-August. The highest cell counts were found during a bloom of the dinoflagellate Prorocentrum minimum in March inside Mobile Bay. The community composition showed a seasonal succession from dinoflagellate- to diatom-dominance in spring, followed by mixed assemblages of chlorophytes, prasinophytes and cryptophytes in summer. There were also horizontal gradients, with dinoflagellate-dominance occurring only in Mobile Bay during the Prorocentrum. There were higher proportions of prasinophytes in Mobile Bay and higher proportions of chlorophytes offshore in the mixed-flagellate assemblage in summer.
- Purpose:
- The purpose of this study is to take a census of the abundance, taxonomic composition and productivity of microalgal populations over an environmental gradient of nutrient and light availability over 3 annual cycles. The shallow coastal northern U.S. Gulf of Mexico (GOM) are physically dynamic and highly productive. Microalgal populations, whose growth supports the productivity of higher trophic levels are variable with respect to composition, abundance and growth potential. Accounting for this variability is essential to development of models of trophic structure and function.

- Completeness_Report:
- Foul weather on the May and November sampling events resulted in the completion of only 10 and 5 of the 15 sites respectively. A malfunction in the winch used for sampling on the event in June prevented the entire sampling regimen to be completed. Due to high winds, three attempts were made in January 2008, however no samples were obtained. The event in March was canceled due to an emergency usage of the E.O. Wilson to look for lost instruments on the missing mooring buoy and was not able to be rescheduled due to lack of vacancy in reserving the vessel. Fowl weather prevented the cruise in mid-May, and it was rescheduled for a half day due to boat availability, therefore only site T20 was successfully sampled. The alternate day in late May was unsuccessful due to instrument communication problems. Fowl weather prevented the sampling of the station T35 and 3 intercalary stations in June, therefore 18 of the 25 sites were collected.

- 1) Flow-through monitoring Synchronize computers to time.gov and, where possible, initialize instruments so that they collect on the minute. For the MIDAS, collect on the minute. For instruments running through the MPak, collect 30 s records centered on the minute. 2) Sampling on station Check and record time on and off station, the GPS co-ordinates, bottom depth, T and S. Run a light cast with the MicroPro, making sure not to lower it deeper than 1 m off bottom. Run a CTD cast with the rosette, reversing when the depth reading is 1 m off bottom. Fire all the Niskin bottles at 1 m off bottom, and collect through Nitex screen into the sample bottles and place on ice in a cooler. Collect and preserve cell samples: one with paraformaldehyde, one with isopropyl and one with Lugols (ADPH's). Cap, mix and place in darkness as soon as possible. Wear gloves when handling the paraformaldehyde and put the samples into a cooler with ice as soon as possible. Place all paraformaldehyde-contaminated waste in a sealed Ziplock. Collect a 2nd and 3rd samples at 2/3 and 1/3 of water depth on the two offshore stations. Collect surface samples at both offshore and inshore stations and sieve through a Nitex screen into a sample bottle. 3) Sampling underway Collect a single sample from the continuous-flow system at each of the intercalary CTD stations. Record the precise collection time (use laptop), to allow match-up with optical data. These samples are for work-up of optics (Chla, aPh, aDet and aCDOM, dissolved nutrients) for ground-truthing.
