Abstract:
Following crude oil contamination from the Deepwater Horizon explosion in June of 2010 of a marsh ecosystem in Alabama, hydrocarbon degrader microbial abundances of aerobic alkane, total hydrocarbon, and polycyclic aromatic hydrocarbon (PAH) degraders were enumerated seasonally. Surface sediment samples were collected in October and December of 2010, and April and July of 2011 along a 40 to 70 m transects from the high tide to the intertidal zone and included Spartina alterniflora vegetated marsh, Ruppia maritima dominated sediments, and non-vegetated sediments. Alkane and total hydrocarbon degraders in the sediment were detected, while PAH degraders were below detection limit at all locations examined during the sampling periods. The highest counts for microbial alkane degraders were observed at the high tide line in April sediment.
Purpose:
The purpose of the study was to determine seasonal changes in microbial communities after the Deepwater Horizon Oil Spill in a Point aux Pines, Alabama marsh ecosystem.

Process_Description:
Samples were collected from locations between 30°22’43” and 30°22’45” North and between 88°18’11” and 88°18’13” West at a Alabaman salt marsh ecosystem at Point Aux Pines (PAP). PAP, located in the northern Mississippi Sound, is dominated by Juncus roemerianus and Spartina alterniflora at its seaward edge and the seagrass (Ruppia maritima) in the sub-tidal zone. Patches of non-vegetated sediments are interspersed between S. alterniflora and R. maritima. Three submerged samples were collected representing mud with well graded sediments, poorly graded sandy sediments with R. maritima vegetation, and poorly graded sandy sediments without vegetation. At the seaward edge of the zone dominated by R. maritima, non-vegetated sandy sediments dominate the landscape. In June 2010, tar balls and oil slicks were detected at this site. Sediment cores (30.3 cm H, 3.5 cm D) were collected seasonally, in October (fall season) and December (winter), 2010 and April (spring) and July (summer), 2011 perpendicular to the shoreline. Sediments were collected at the marsh dominated by S. alterniflora, subtidal zone dominated by R. maritima, and non-vegetated sediments. In October, 2010, April and July, 2011 cores were collected at high tide line (HTL) in a high density S. alterniflora plot; 5 m away from HTL with less abundant S. alterniflora; at 10 m from the HTL from a non-vegetated muddy area; at 20 m from the HTL in a R. maritima dominated sediments; and at 40 m away from HTL in a non-vegetated and sandy sediment zone. At the time of sampling marsh sediments were exposed to air, while all other cores were submerged during the collection. In December 2010, the cores were collected further apart due to lower tides but still represented patches similar in composition as those collected in other months. Surface sediments were collected from the top 2 cm of the cores, and 9 to 11 cm interval from the sediment surface. In January and July, 2011, the top 15 cm of sediments were collected for hydrocarbon analyses at each sampling location along the transects.
